There's a little less high end in the covered pickup part of the demo, and a bit less crunch due to the reduction of treble (people used to use treble boosters to get more crunch out of their guitars). There's also a bit more clarity in the first pass of the first covered pickup (don't remember if it was the neck or bridge pickup).
I was able to hear the difference quite clearly on my macbook pro's built-in speakers, I didn't bother going into the studio to listen since it was so noticeable.
Of course, this isn't a demo of 408s, but the main distinction in the 408 split coil sound is a bit more attack in the high end of each note, and a little bit more clarity than the humbucker tones.
So if the 408s respond to being covered like most pickups, it's going to be much as I said.
